Monkeypox viruses remain sensitive to the available drugs

 The mpox virus is closely related to the smallpox virus (variola virus), which caused large, deadly outbreaks before it was eradicated by vaccination at the end of the 1970s. While the smallpox virus led to very severe disease progression with a death rate of about 30%, mpox is milder. Nevertheless, the mortality rate is still about 3%.

Particularly at risk of a severe course of the disease are people with a weakened immune system, elderly persons, , newborn babies and young children. Until recently, mpox outbreaks only occurred in certain parts of Africa when humans became infected through contact with , typically rodents such as the Gambian pouched rat and the rope squirrel.

However, in May 2022 a first large mpox outbreak outside Africa was detected; the  spread solely through human-to-human transmission. This ongoing outbreak has so far reached more than 100 countries and been classified by the World Health Organization (WHO) as a Public Health Emergency of International Concern.

About 10% of mpox patients require hospital treatment. Moreover, the current mpox outbreak differs from previous ones in terms of both disease transmission and symptoms. These differences raised concerns that the currently circulating mpox virus might have changed in such a way that it would no longer respond to the  available.

Against this backdrop, an international research team led by Professor Jindrich Cinatl from the Institute of Medical Virology, Goethe University Frankfurt/University Hospital Frankfurt, and Professor Martin Michaelis from the School of Biosciences at the University of Kent have succeeded in isolating and cultivating viruses in cell culture from 12 patients from the current mpox outbreak. This has enabled them to test these mpox virus isolates in cultures of skin cells, which has been naturally infected by the mpox virus, for their sensitivity to three drugs presently available to treat the disease: tecovirimat, cidofovir and brincidofovir.

The results showed that all 12 isolates continued to respond to treatment with clinically relevant concentrations of these commonly used drugs.

Professor Jindrich Cinatl said, "We were really concerned that the virus could have changed and become resistant to the available therapies. It is good to see that this is not the case."

Professor Martin Michaelis added, "These findings are very reassuring and give good cause to believe that the antiviral drugs already available will also be effective against the mpox virus in the current outbreak."

Machine learning technique on the hypothalamus gives insight into nature of aggression

 Have you ever been cut off while driving and found yourself swearing and laying on the horn? Or come home from a long day at work and lashed out at whoever left the dishes unwashed? From petty anger to the devastating violence we see in the news, acts of aggression can be difficult to comprehend. Research has yielded puzzling paradoxes about how rage works in the brain. But a new study from Caltech, pioneering a machine-learning research technique in the hypothalamus, reveals unexpected answers on the nature of aggression.

The hypothalamus is a  region linked to many innate survival behaviors like mating, hunting, and the fight-or-flight response. Scientists have long believed that neurons in the hypothalamus are functionally specific—that is, certain groups of neurons correlate to certain specific behaviors. This seems to be the case in mating behavior, where neuron groups in the medial preoptic area (MPOA) of the hypothalamus, when stimulated, cause a  to mount a female mouse. These same neurons are active when mounting behavior occurs naturally. The logical conclusion is that these neurons control mounting in mice.

But when looking at the analogous neurons that control  in another part of the hypothalamus, the VMHvl, researchers found a different story. These neurons could be stimulated to cause a male mouse to attack another male mouse, yet they did not show specific activity when the same neurons were observed in naturally fighting mice. This paradox indicated that something distinct was happening when it came to aggression.

Now, a new Caltech study provides insight into the unique neural mechanisms of anger and shows that more aggressive dispositions are codified in the mouse brain's circuit dynamics. The research was published in the journal Cell on January 5 and was conducted in the laboratory of David Anderson, the Seymour Benzer Professor of Biology, Tianqiao and Chrissy Chen Institute for Neuroscience Leadership Chair, a Howard Hughes Medical Institute Investigator, and director of the Tianqiao and Chrissy Chen Institute for Neuroscience. The study is part of a continuing collaboration with Ann Kennedy, a theoretical neuroscientist and former postdoctoral fellow in the Anderson lab, now an assistant professor of neuroscience at Northwestern University.

The study's lead author, Caltech graduate student Aditya Nair, applied a  to analyze  and uncover hidden patterns in .

"We used machine learning to fit a dynamical system model to recorded neural data from mice," Nair says. "Essentially, the dynamical system acts like an artificial mouse brain in a computer which reflects our experimental observations. We then studied this model in depth to learn how these hundreds of neurons talk to one another. By applying dimensional reduction and other analysis techniques from physics, we were able to finally see the neural signals and computations causing aggression."

The analysis revealed a "line attractor" in the brain—a mathematical representation of a pattern that emerges from complex neural systems. When graphed in an energy landscape, the line attractor forms the shape of a trough. Though the trough is not a  in the brain, it indicates the flow of energy happening in the dynamical system.

"Discovering the line attractor was exciting because it showed for the first time a neural correlate to aggressive escalation, a key finding with important societal implications," Nair explains.

Mice typically show  when another male mouse is introduced to their cage during experiments. Their behavior escalates from threatening displays to full-blown attack, a sliding-scale trajectory that can now be explained by line-attractor dynamics. As neural activity progresses along the line-attractor trough, the mouse's aggression ramps up. The shape of the trough makes it simple for the brain to modulate aggression—like a volume dial that can be turned up or down. If the "volume" gets too high, animals attack. This modulation may serve as an evolutionary adaptation, compelling animals to measure out displays of aggression and test the waters before going into a potentially deadly fight.

The discovery of a line attractor was also, unexpectedly, correlated with individual differences in aggression. The study's dynamical system model showed that mice that became angry and attacked more quickly had deeper line-attractor troughs, indicating that aggressive temperaments are reflected in the brain's structure. Because these lab mice are genetically identical, these individual differences must be due to epigenetic factors, the nature of which remain to be discovered.

Though attractor dynamics have been discovered in "higher" brain regions such as the cortex or hippocampus, often linked to  and memory, this study is one of the first to use a dynamical systems approach to understand social behaviors in the deep brain.

Could attractor dynamics be the driving force behind other internal emotional states? Do we control our emotions, or do they control us? The findings in this study evoke questions and future directions for research, open the door to therapeutic drug development for emotional disorders, and elevate our understanding of how the hypothalamus encodes aggression.

DeSantis Activates National Guard Amid Surge of Illegal Aliens Entering Florida Keys

Florida Gov. Ron DeSantis on Friday signed an executive order to activate the National Guard in the state amid a major surge of illegal aliens arriving in the Florida Keys from places such as Cuba and other Caribbean countries.

“As the negative impacts of [President Joe] Biden’s lawless immigration policies continue unabated, the burden of the Biden administration’s failure falls on local law enforcement who lack the resources to deal with the crisis,” the Republican governor said in a statement.

“That is why I am activating the National Guard and directing state resources to help alleviate the strain on local resources. When Biden continues to ignore his legal responsibilities, we will step in to support our communities.”

The executive order (pdf) also directs state law enforcement agencies and other state agencies to “provide resources in support of local governments” responding to the influx of illegal immigrants in the Florida Keys, as well as “additional support toward efforts to prevent further migrant landings on Florida’s shores.”

Florida’s state government is also set to deploy aircraft from the Florida National Guard, as well as support marine patrol from the Florida Fish and Wildlife Conservation Commission, to “support water interdictions and ensure the safety of migrants attempting to reach Florida through the Florida Straits.”

DeSantis’s office called the influx of illegal immigrants “alarming.”

It noted that about 300 illegal aliens on Jan. 1 unlawfully entered Dry Tortugas National Park, which is about 70 miles west of Key West. On the same day, another 45 illegal aliens entered Key West—an island that’s part of the Florida Keys archipelago.

“This massive influx of illegal immigrants caused the park to close to ensure the safety of visitors and staff and to provide the space necessary to attend to the illegal immigrants,” stated the governor’s office.

“Since August 2022, federal, state and local law enforcement have encountered more than 8,000 migrants in waters off the coast of Florida.”

“Florida has a long history of helping refugees, including Cubans and others fleeing communist regimes, find support after they arrive in the United States, however, this has always involved support from the Federal Government and a large, coordinated effort amongst state, federal and local governments,” DeSantis’s office said.

But it is “particularly burdensome” for Monroe County Sheriff’s Office, which has 194 deputies, to manage hundreds of illegal immigrants while also trying to ensure public safety, his office noted. It called the Biden administration’s overall response to the influx of illegal aliens into the United States “inept,” adding that the federal government has failed to “provide the resources necessary to respond to the current mass migration event.”

“Instead, the burden falls on local law enforcement, which puts Floridians at risk.”

Meanwhile, the arrivals have been continuing.

Walter N. Slosar, the chief patrol agent of the Miami Sector headquarters, wrote on Twitter late Jan. 6: “During the past 24 hours, Border Patrol agents & [law enforcement] partners responded to 5 migrant landings & encountered 90 Cuban migrants in the Florida Keys. The migrants arrived on rustic vessels & residents notified local authorities.”

Twitter further cuts staff overseeing global content moderation

 

Twitter Inc made further staff cuts in the trust and safety team handling global content moderation and in the unit related to hate speech and harassment, Bloomberg news reported on Saturday.

At least a dozen more cuts on Friday night affected workers in the company's Dublin and Singapore offices, the report said, citing people familiar with the matter.

Twitter, owned by billionaire Elon Musk, did not immediately respond to a Reuters request for comment.

NY nurses reach deals with some hospitals, others still plan strike

 

  • Some of the thousands of nurses planning to strike at New York hospitals Monday have reached a deal with their facilities to avert the planned open-ended strike, according to union officials.
  • Nurses at three facilities: New-York Presbyterian, Maimonides and Richmond University medical centers, made tentative agreements averting strikes, while nurses at five other hospitals are still bargaining for new contracts.
  • Those still planning to strike want contract terms that ensure adequate staffing and address widespread turnover and burnout driven by the COVID-19 pandemic, according to a release from the New York State Nurses Association.
  • Nurses at BronxCare Health System, Flushing Hospital Medical Center, Montefiore Bronx, Mount Sinai Hospital and Mount Sinai Morningside and West are still slated to walk off the job Monday if they can’t reach a deal with the systems on new contracts by then.

Some 16,000 nurses initially voted to strike without a set end date, delivering official 10-day notice to the systems just before contracts expired.

On Dec. 30, 4,000 nurses at NewYork-Presbyterian reached a tentative agreement with the system on new contracts, though, halting a strike there.

That deal includes raises of 7% in the first year of the contract, 6% in the second and 5% in the third, along with other terms to improve staffing, union leaders said on a call with reporters Thursday.

On Wednesday, nurses at Maimonides and Richmond University medical centers also reached deals, though details of those contracts have not yet been released.

Exact contract terms also vary by facility, union leaders said.

Montefiore Senior Vice President Joe Solmonese said in a statement the hospital proposed contract terms similar to those in the deal at New-York Presbyterian, including 18% wage increases over the course of the contract and other benefits that nurses will not accept.

Union leaders on the call pushed back and said staffing is still a priority in negotiations.

New York passed a staffing law last year requiring hospitals and nursing homes to form clinical committees tasked with setting annual staffing standards for units. NYSNA leaders on the call said implementation and enforcement is still spotty, however, and the union is now pushing for a minimum standard throughout the state.

Ontario Governing Body For Psychologists Demands Re-Education For Jordan Peterson

 Outspoken psychologist and former tenured professor Jordan Peterson has come under relentless attack ever since he criticized the 'Act To Amend The Canadian Human Rights Act And The Criminal Code' back in 2016.  The act is a framework which makes scrutiny of gender identity ideology or transsexualism into potential discrimination under Canadian law.  Peterson posted a series of videos which outline why critique of gender identity groups is rooted in science and should be protected as free speech.    

After several years of harassment from leftist activists and politically motivated bureaucrats, Peterson is once again being threatened.  This time, with the loss of his license to practice psychology unless he submits to a series of 're-education' programs mandated by the College of Pyschologists of Ontario.

The CPO is a quasi-governmental body (a mostly unaccountable bureaucracy) acting in accordance with the Psychology Act, 1991 the Health Professions Procedural Code, the Regulated Health Professions Act, 1991 and the regulations and by-laws of the college.  The CPO's sudden aggressive effort to silence Jordan Peterson appears to have been triggered by his reinstatement on Twitter in November.

He was originally banned from the site for referring to actress Elliot Page by her original name (Ellen Page) and using the word “her” as a descriptive, which is a scientific fact, but also a violation of the leftist site's TOS before Elon Musk's takeover.  

The psychologist is also known for his criticisms of Canada's draconian covid lockdowns and mandates.

With Peterson once again having access to a wider audience on social media, the CPO has ordered him to complete a mandatory “Specified Continuing Education or Remedial Program” to “review, reflect on and ameliorate [his] professionalism in public statements,” according to a lengthy list of requirements from the college that Peterson shared on Twitter.  He must meet with a psychologist for coaching classes, which he must pay for, until a final report is issued by the coach that shows their concerns have been “properly ameliorated.”    

The CPO argues that Peterson's social media interactions constitute a violation of the organization's standards for professional interactions with the public.  In other words, the CPO essentially believes it has the right to control the speech of member psychologists and force them to adhere to a predetermined narrative they find acceptable when engaging with the public.  

It should be noted that there are no publicized instances of psychologists being threatened with a loss of their license for espousing leftist viewpoints on social media.  The measures appear to be strictly designed to punish more conservative leaning professionals.  

Peterson says he will not comply with the CPO programs because his comments have nothing to do with his role as a psychologist.  “Every single accusation is not only independent of my clinical practice, but explicitly political — and not only that: unidirectionally explicitly political,” Peterson writes.

Canada's spiral into authoritarianism is widely publicized and the nation's waning respect for free speech accelerated during the pandemic lockdowns.  While the US is not far behind, Americans at least have free speech rights codified in a constitution that is designed to regulate what the government can and cannot do.  In Canada, such a thing does not exist.  Canadian rights are treated more like privileges that the government is allowed to legally erase at a moment's notice.

CPO actions not only indicate hostility towards free speech, but also hint at an underlying fear – Members of the professional class are often the most indoctrinated of all people, and for one of them to break from the paradigm and speak against the establishment is considered the utmost betrayal.  

Peterson is being targeted with a special brand of bureaucratic attention because the establishment uses the professional class to convince and control the general public.  The claim that the “experts have spoken” is the ultimate tool for manipulating a majority of the population, and when some of those experts contradict the narrative, the powers-that-be take particular notice.

Tesla vehicles 40% cheaper in China than US after major price cuts

 Tesla has slashed prices on some vehicles in China by more than 40% compared to their sticker price in the US as Elon Musk’s struggling electric automaker responds to dwindling demand in the region.

After a fresh round of discounts unveiled this week, the base version of Tesla’s Model Y SUV is available for the equivalent of $37,875 in China – a 43% discount compared to the cheapest available Model Y on sale in the US, Bloomberg reported Friday.  The new price in China marked a 10% discount from its previous level.

Tesla also chopped its ask for the base version of its flagship Model 3 in China by 10% to the equivalent of about $33,575. The Model 3 is currently about 30% more expensive in the US.

Tesla also cut prices for vehicles in Japan, South Korea and Australia as it attempts to boost demand and head off increased competition in the electric vehicle market. The company had previously lowered prices as recently as October.

The latest cuts were unveiled days after Tesla spooked investors by reporting fourth-quarter vehicle delivery numbers that fell short of Wall Street’s expectations. The 405,278 vehicle deliveries were an internal record but was still below the 432,117 that analysts expected.

Despite the news, Tesla shares rose 2.5% on Friday. The company’s stock is off to a rough start this year after posting a 65% decline in 2022 – its worst annual performance on record.

In a post on the Chinese social media platform Weibo, Tesla executive Grace Tao said the discounts were the result of engineering innovations and calls from Beijing to boost economic development.

Tesla executives have grappled with months of supply chain difficulties, especially in China, where a surge in COVID-19 cases forced the company to temporarily shutter production at a key manufacturing plant in Shanghai. Production is expected to be limited through January.

Elsewhere, Tesla investors have grown irritated as Musk devotes more of his time and energy to running Twitter. Analysts say Musk’s controversial moves at Twitter, including rounds of layoffs, cost-cutting measures and an overhaul of the site’s verification process, have become an overhang on Tesla’s stock.

Last week, several Wall Street firms cut their price targets for Tesla in response to the lukewarm delivery report. Tesla’s quarterly numbers are closely watched because they are the closest indication of actual unit sales disclosed by the company.

“Demand overall is starting to crack a bit for Tesla and the company will need to adjust and cut prices more, especially in China which remains the key to the growth story,” Wedbush analyst Daniel Ives said in a note to clients.

The price of used Tesla vehicles has also begun to fall, plunging 17% from a peak of $67,297 in July to $55,754 in November, according to a recent Reuters report. Over the same span, the overall price of used cars fell just 4%.
